Abstract: Solanum betaceum Cav., vulgarmente conhecida como tamarilho, é uma pequena árvore andina pertencente à família das solanáceas, cultivada pelos seus frutos comestíveis. Esta espécie tem sido estudada devido ao seu particular interesse na compreensão de aspetos da morfogénese in vitro, em processos como a embriogénese somática. Atualmente, a embriogénese somática é uma ferramenta de alto valor biotecnológico para a clonagem em larga escala e um interessante modelo para o estudo de processos que ocorrem durante o desenvolvimento embrionário. Diferentes aspetos relacionados com a embriogénese somática no tamarilho têm sido estudados no nosso laboratório, mostrando diferentes respostas morfogénicas dependendo do tipo de frasco de cultura usado. A causa destas diferenças parece ser influenciada pela atmosfera de cultura, que não é apenas constituída por gases atmosféricos, mas também por etileno. O etileno é uma hormona vegetal que tem sido relacionada com processos de morfogénese in vitro e por essa razão é importante perceber se é o componente da atmosfera de cultura responsável por estas diferenças morfológicas. Para verificar esta hipótese, foram realizados diferentes ensaios relacionados com este processo, como a indução da embriogénese somática, a proliferação de calo embriogénico e não embriogénico e a formação de embriões somáticos. A resposta desta espécie em relação a estes ensaios foi testada em diferentes meios MS contendo 50 µM de nitrato de prata (AgNO3), 10 µM de aminoethoxivinilglicina (AVG) e 20 µM ethephon (ETH). Durante estes ensaios foi também tentado quantificar a concentração de etileno nos frascos de cultura. Na fase de indução, os explantes foliares que foram expostos a AgNO3 e AVG apenas produziram calo não embriogénico e na presença de ETH verificou-se um aumentou significativo da indução de tecido embriogénico. Além do mais, no tratamento com ETH, foi observada a formação de embriões somáticos ao fim da oitava semana no meio de indução. Estes resultados sugerem que o etileno tem um papel fundamental na aquisição da competência embriogénica no tamarillo assim como aparenta potenciar a formação de embriões somáticos através de um processo “one-step”, revertendo o efeito inibitório das auxinas no meio de cultura. A sobre-exposição ao etileno parece ter um efeito negativo na proliferação de calo embriogénico, mas não parece afetar a proliferação de calo não embriogénico. Na fase de desenvolvimento, o tratamento com AgNO3 e AVG aumentou o número de embriões somáticos desenvolvidos a partir de calo embriogénico, enquanto na presença de ETH a sua formação foi inibida. Estes resultados indicam que uma menor ação de etileno em tecido embriogénico de tamarilho parece promover a formação de embriões somáticos num meio sem auxinas. Apesar dos mecanismos moleculares da ação do etileno neste estudo ainda não sejam conhecidos, no seu conjunto, estes resultados sugerem que o etileno tem um papel específico em diferentes estágios da embriogénese somática no tamarilho, que podem estar relacionados com uma possível interação com auxinas. Este estudo é uma primeira abordagem para a compreensão do efeito do etileno na embriogénese somática do tamarilho, assim como, um ponto de partida para uma melhor caracterização dos aspetos moleculares envolvidos neste processo.
Solanum betaceum Cav., commonly known as tamarillo, is an Andean small solanaceous tree, which is cultivated for its edible fruits. This species has been studied due to its particular interest in understanding aspects of in vitro morphogenesis, in processes such as somatic embryogenesis. Currently, somatic embryogenesis is a high-value biotechnological tool for large-scale cloning and an interesting model to study processes that occur during embryonic development. Different aspects related to somatic embryogenesis in tamarillo have been studied in our laboratory, showing different morphogenic responses depending on the type of culture vessel used. The cause of these differences seems to be influenced by the culture atmosphere, which is constituted not only by atmospheric gases, but also by ethylene. Ethylene is a plant hormone that has been related to in vitro morphogenesis processes and for that reason it is important to determine whether ethylene is the component of the culture atmosphere responsible for the morphological differences observed. To verify our hypothesis we have carried out different assays related to somatic embryogenesis, such as somatic embryogenesis induction, proliferation of embryogenic and non-embryogenic callus and somatic embryo formation. The response of tamarillo with regard to these diverse essays was tested on different MS medium containing 50 µM silver nitrate (AgNO3), 10 µM aminoethoxyvinylglycine (AVG) and 20 µM ethephon (ETH). During these essays, it was also tried to quantify ethylene concentration in the culture vessels. At the induction stage, the leaf explants exposed to AgNO3 and AVG have only produced non-embryogenic callus and the presence of ETH increased significantly the induction of embryogenic tissue. Furthermore, in ETH treatment, the formation of somatic embryos was observed after 8 weeks in the induction medium. These results suggests that ethylene plays a fundamental role in the acquisition of embryogenic competence in tamarillo as well as an apparent potentiation of somatic embryo formation through a one-step process, reversing the inhibitory effect of auxins on the culture medium. The overexposure of ethylene appears to be negative for embryogenic callus proliferation, but does not have effect on non-embryogenic callus proliferation. At the development stage, the treatment with AgNO3 and AVG has enhanced the number of somatic embryos developed from embryogenic callus while the presence of ETH inhibit their formation. These results indicate that a lower action of ethylene in embryogenic tissue of tamarillo seems to promote the formation of somatic embryos in a medium without auxins.Although the molecular mechanisms of ethylene action in this study remain unknown, taken together these results suggest that ethylene plays a specific role at the different stages of SE in tamarillo, which may be related to a possible interaction with auxins. This study is a first approach to clarify the effect of ethylene on somatic embryogenesis of tamarillo as well as a starting point for a better characterization of the molecular aspects involved in this process.
