betaceum, uma solanácea filogeneticamente próxima do tomateiro (Solanum
lycopersicum), levaram à descoberta de uma SpoU metiltransferase de RNA - NEP-TC
(Non-Embryogenic Protein from Tamarillo Callus). Esta proteína poderá ter um eventual
papel como regulador negativo da embriogénese. Em Arabidopsis thaliana, a
superfamília de SpoU metiltransferases inclui pelo menos cinco proteínas, codificadas
por diferentes genes, que partilham o domínio conservado SpoU MeTrfase (IPR001537).
Este trabalho teve como objectivo avaliar a função de algumas destas proteínas ao analisar
características fenotípicas e as taxas de indução de embriogénese somática em sementes
wild type e sementes knockout (linhas mutantes SALK) de Arabidopsis thaliana, para os
genes codificantes destas metiltransferases e através da análise de linhas de plantas de
tamarilho silenciadas para o gene codificante da NEP-TC.
Para analisar os parâmetros fenotípicos todas as plantas foram cultivadas em turfa
com vermiculite ou em cultura in vitro em em meio MS. Os parâmetros medidos em A.
thaliana, foram o comprimento das raízes primárias e o número de raízes secundárias,
bem como outros aspetos do desenvolvimento, incluindo o aparecimento das primeiras
folhas da roseta ou da primeira síliqua. Em Solanum betaceum, os parâmetros medidos
foram o número e o comprimento das raízes adventícias e o número de entrenós de plantas
micropropagadas. No que diz respeito à análise das taxas de embriogénese somática em
A. thaliana todas as sementes foram colocadas em cultura em meio MS suplementado
com 0,1 mg/L de 2,4-D. Para o tamarilho foram colocados segmentos foliares em meio
MS suplementado com 5,0 mg/L de picloram.
Em termos do enraizamento, os resultados obtidos para as linhas de A. thaliana
foram muito semelhantes no que diz respeito ao comprimento das raízes primárias entreas linhas mutantes e a wild-type, verificando-se diferenças significativas no número de
raízes secundárias, com um número muito superior nas linhas mutantes
comparativamente com a linha wild type. Em relação às taxas de desenvolvimento, os
resultados obtidos para as linhas mutantes foram muito similares, apresentando estas no
geral um atraso em relação à linha wild type. Ao nível da embriogénese somática as linhas
mutantes apresentaram taxas de indução mais elevadas comparativamente com a wild
type, com diferenças de 30% nas taxas de indução.
Na análise das linhas de tamarilho silenciadas para o gene codificante da NEPTC,
ao nível do enraizamento apenas se registaram diferenças significativas no
comprimento da raiz primária para uma das linhas transformadas. No que diz respeito ao
número de raízes e de entrenós os resultados foram muito semelhantes não havendo
diferenças significativas. Ao nível das taxas de indução de embriogénese somática os
resultados foram muito díspares e não possíveis de relacionar com os resultados da linha
controlo, obtendo-se taxas de indução de embriogénese somática na ordem dos 35% para
uma das linhas silenciadas.
Estes resultados permitem concluir que estes genes e as proteínas que codificam
têm um efeito significativo no processo de embriogénese somática actuando como
reguladores negativos tanto em Arabidopsis thaliana como em Solanum betaceum. No
que diz respeito ao desenvolvimento das plantas também aqui o efeito parece ser muito
significativo, particularmente ao nível do desenvolvimento radicular. Mais
especificamente ao nível das raízes secundárias em A. thaliana e no comprimento das
raízes adventícias em tamarilho. Previous studies in Solanum betaceum somatic embryogenesis, a solanaceae closely related to tomato (Solanum lycopersicum), led to the identification of a putative SpoU methyltransferase - NEP-TC (Non-Embryogenic Protein from Tamarillo Callus), systematically expressed on non-embryogenic tissues. In Arabidopsis thaliana, the SpoU rRNA Methylase family family includes at least five different proteins, expressed by different genes, that share the conserved domain SpoU_MeTrfase (IPR001537). The objective of this work was to evaluate the function of some of these proteins on plant development, particularly in the induction of somatic embryogenesis on wild type seeds vs knockout seeds (SALK mutant lines) for the genes encoding those proteins, and also in Solanum betaceum silenced lines for the gene encoding NEP-TC. To measure the phenotypic parameters all plants were grown in turf with vermiculite or micropropagated in MS medium. The parameters measured for Arabidopsis thaliana were the length of the primary roots and the number of secondary roots, as well as the time of some development stages, such as time of germination, appearance of first rosette leaves or appearance of first silique. In Solanum betaceum, the parameters measured were the number and length of adventitious roots and the number of internodes in micropropagated plants. To analyse the somatic embryogenic induction rates in A. thaliana, all seeds were cultured in MS medium supplemented with 0,1 mg/L 2,4-D. To analyse the induction rates in S. betaceum, leaf segments were cultured in MS medium supplemented with 5,0 mg/L of picloram. The results showed that in A. thaliana, the length of the primary roots were very similar. However, in terms of the number of secondary roots the mutant lines showed a higher number on average. On the development rates the results were very similar amongst all mutant lines with a small delay when compared with the wild type. Regardingthe somatic embryogenesis induction rates, the mutant lines showed higher induction rates when compared to the wild type line, with the differences reaching 39%. With regards to Solanum betaceum, the results in rooting analysis showed differences in the length of the adventitious roots for one line which showed a higher length with significant differences to all others. As far as the number of roots and number of internodes there was no significant difference in any of the lines. As for the somatic embryogenesis induction rates there were very despair results, not possible to compare with the control line, but with induction of embyogenic callus reaching percentages of 35%. Concluding, these genes and the proteins they encode seem to have a significant effect in the process of somatic embryogenesis acting as a negative regulators both in Arabidopsis thaliana and Solanum betaceum. As far as influencing the phenotype of the plants, once again in both species they seem to have a pretty significant effect on the roots. More specifically on the number secondary roots in A. thaliana and the length of adventitious roots in tamarilho. |
