existentes, na qual a sua estrutura ainda permanece largamente desconhecida, sendo um dos
grandes objetivos mais desafiadoresde esclarecer. Na sua composição existem diversos
componentes como neurónios, células glias, vasos sanguíneos que formam redes
neurovasculares extremamente complexassendo fundamental para as funções neurais
avançadas. Para o proteger e manter a homeostase cerebral existe um sistema biológico
altamente seletivo denominado de barreira hematoencefálica (BHE). Na composição da BHE
existem quatro elementos celulares fundamentais: células endoteliais, astrócitos, pericitos e
células da microglia permitindo assim que esta possua uma serie de características
fundamentais como a sua impermeabilidade e seletividade. A sua disfunção provoca uma
alteração da homeostasia cerebral podendo levar a progressão de várias doenças do Sistema
Nervoso Central (SNC).
Os tumores cerebrais afetam cada vez mais a população em todo mundo a cada ano,
sendo um dos principais problemas de saúde pública podendo ter efeitos letais. Atualmente
um dos grandes objetivos é provocar a abertura da BHE para que se seja possível tratar este
tipo de tumores evitando recorrer à cirurgia e eventualmente a tratamentos pouco
específicos. Inicialmente eram utilizados sistemas convencionais não específicos para
permeabilizar a BHE, mas estes tinham uma limitação, a falta de alvos específicos na BHE.
Assim com o aparecimento da Técnica de Focagem de Ultrassons (FU) juntamente com as
microbolhas, é possível provocar a abertura da BHE cerca de 4-6 horas, de uma forma não
invasiva e reversível com a aplicação transcraniana de ondas de ultrassom de baixa
frequência, resultando assim numa entrega mais eficiente de agentes terapêuticos nas
diferentes regiões cerebrais.
A FU permite realizar a entrega cerebral de anticorpos, agentes quimioterápicos,
nanopartículas e genes específicos, que permite tratar doenças neurodegenerativas e
essencialmente diferentes tumores cerebrais, na qual representa uma grande promessa no

Regarding composition, there are several components such as neurons, glia cells, blood vessels that form extremely complex neurovascular network which is central to the advanced neural functions. To protect and maintain brain homeostasis there is a highly selective biological system called blood-brain barrier (BBB). The composition of the BBB are four fundamental cellular elements: endothelial cells, astrocytes, pericytes and microglial cells thus allowing this has a number of key features as its waterproof and its selectivity. Its dysfunction causes a change of brain homeostasis and can lead to the progression of various diseases of the central nervous system (CNS). Brain tumors affect more and more people worldwide each year, becamingone of the main public health problems that can have lethal effects. Currently one of the major goals is to induce the opening of the BBB so that it is possible to treat such tumors avoiding resorting to surgery and eventually some specific treatments. Initially, nonspecific conventional systems to permeabilize the BBB were used, but these had a limitation, the lack of specific targets in the BBB. Thus with the onset of Focused Ultrasound Technique (FU) with the microbubbles, it is possible to cause the opening of the BBB for about 4-6 hours, in a non invasive and reversible manner with the transcranial applying of low frequency ultrasound, thus resulting in a more efficient delivery of therapeutic agents in different brain regions. FU allows brain delivery of antibodies, chemotherapeutic agents, nanoparticles and specific genes which allow treating neurodegenerative diseases and brain tumors primarily different, which holds great promise for the future |
