A terapia fotodinâmica é um procedimento clinicamente aprovado que tem demonstrado um grande potencial para o tratamento de tumores sólidos. Esta terapia é caracterizada pela administração de um fotossensibilizador que quando irradiado, na presença de oxigénio, vai gerar espécies reativas de oxigénio. Estas moléculas têm a capacidade de criar um stress oxidativo que vai danificar as biomoléculas envolventes e consequentemente, pode causar a morte das células tumorais. A terapia fotodinâmica, para além da ação directa que tem no cancro, apresenta ainda a vantagem de estimular o sistema imunitário a reconhecer e destruir células cancerígenas. Os lípidos são um dos tipos de biomoléculas que podem sofrem oxidação após tratamentos de terapia fotodinâmica. Os lipídios podem ser oxidados pelas espécies reactivas de oxigénio ou diretamente pelo fotossensibilizador no estado tripleto e como resultado, surgem uma série de moléculas, algumas das quais com propriedades citotóxicas. Tipicamente, após a oxidação lipídica é característico observar-se a formação de malonaldeído, 4-hidroxi-2-nonenal, hidroperóxidos lipídicos assim como a diminuição dos tióis. Estas moléculas podem ter diversos efeitos biológicos como por exemplo, a ativação de diversas vias de sinalização associadas a variados mecanismos de morte celular. Este trabalho teve como objectivo perceber a extensão da oxidação lipídica causada por tratamentos de terapia fotodinâmica usando três fotossensibilizadores com lipofilicidades diferentes: LUZ51b, LUZ 11α3β e LUZ10. Num primeiro conjunto de estudos, optimizaram-se as concentrações e doses de luz necessárias de modo a obter níveis de morte celular semelhantes para os três fotossensibilizadores. Estas condições foram usadas para os estudos posteriores relacionados com a avaliação lipídica. Os resultados obtidos nos nossos estudos exploratórios demonstraram que a terapia fotodinâmica com as três moléculas em estudo resultou na produção de espécies reactivas de oxigénio, quer em solução quer no meio intracelular. A avaliação da oxidação lipídica, nomeadamente pela quantificação de hidroperóxidos lipídicos, MDA e grupos tióis demonstra que de facto ocorre a oxidação de lípidos. Contudo, e ao contrário do antecipado, a terapia fotodinâmica com LUZ10 induz maiores níveis de oxidação lipídica. Cancer is one of the leading causes of death worldwide. These molecules can create an oxidative stress that will damage the surrounding biomolecules and consequently can cause the death of tumor cells. Photodynamic therapy, besides its direct action on cancer, also has the advantage of stimulating the immune system to recognize and destroy cancer cells.Lipids are one of the types of biomolecules that can suffer oxidation after photodynamic therapy treatments. Lipids can be oxidized by reactive oxygen species or directly by the photosensitizer in the triplet state and as a result, several molecules emerge, some of which have cytotoxic properties. Typically, after lipid oxidation it is characteristic to observe the formation of malonaldehyde, 4-hydroxy-2-nonenal, lipid hydroperoxides as well as the decrease of thiols. These molecules can have various biological effects such as the activation of various signaling pathways associated with various cell death mechanisms. This work aimed to understand the extent of lipid oxidation caused by photodynamic therapy treatments using three photosensitizers with different lipophilicities: LUZ51b, LUZ 11α3β and LUZ10. In a first set of studies, the required light concentrations and doses were optimized to obtain similar levels of cell death for the three photosensitizers. These conditions were used for the subsequent studies related to lipid evaluation. The results obtained in our exploratory studies demonstrated that photodynamic therapy with the three molecules under study resulted in the production of reactive oxygen species, both in solution and in the intracellular medium. The evaluation of lipid oxidation, namely by quantification of lipid hydroperoxides, MDA and thiol groups demonstrates that lipid oxidation does indeed occur. However, and contrary to what was anticipated, photodynamic therapy with LUZ10 induces higher levels of lipid oxidation. |
