dc.description.abstract | Polyglutamine diseases are a group of nine neurodegenerative diseases caused by the abnormal expansion of the CAG trinucleotide in the coding region of the gene, which originates a mutant protein with a typical segment of polyglutamines. The mutant protein becomes toxic and it leads to progressive neuronal dysfunction and death that gradually limits patients’ lives. Polyglutamine diseases include Huntington's Disease, Dentatorubral Pallidoluysian Atrophy, Spinal and Bulbar Muscular Atrophy, and Spinocerebellar Ataxias type 1, 2, 3, 6, 7, and 17. There is no effective treatment for these diseases or therapeutic alternatives that can limit their progression yet. However, the monogenic nature of polyglutamine diseases makes them ideal candidates for the development of gene therapies. Furthermore, the identification of single nucleotide polymorphisms that are associated with the expanded repeats in each of these diseases allows to selectively target the mutant allele. Therefore, it is possible to preservethe normal allele and the levels of the native protein for an adequate cell performance by genetic silencing of the mutant allele. The main objectives of this monograph were to identify the single nucleotide polymorphisms that are associated with the expanded repeats, as well as the specific allele silencing therapies already studied for each polyglutamine diseases. This research was done by using bibliographical research and genetic databases. dc.description.abstract | As doenças de poliglutaminas consistem num conjunto de nove doenças neurodegenerativas, causadas pela expansão anómala do trinucleótido CAG na região codificante do gene, que dá origem a uma proteína mutante com um segmento característico de poliglutaminas. A proteína mutante torna-se tóxica e, frequentemente, origina disfunção e morte neuronal progressiva que vai limitando progressivamente a vida dos doentes. As doenças de poliglutaminas incluem a Doença de Huntington, a Atrofia Dentato-Rubro-Pálido-Luisiana, a Atrofia Muscular Bulboespinal e as Ataxias Espinocerebelosas tipo 1, 2, 3, 6, 7 e 17. Atualmente não existe tratamento eficaz para estas doenças, nem alternativas terapêuticas que possam limitar a sua progressão. No entanto, o caráter monogénico das doenças de poliglutaminas torna-as candidatas ideais para o desenvolvimento de terapias génicas. Para além, disso, a identificação de polimorfismos de nucleótido único que se encontram associados às repetições expandidas, em cada uma destas doenças, permite alcançar de forma seletiva o alelo mutante. Desta forma, recorrendo ao silenciamento genético do alelo mutante é possível manter o alelo normal e os níveis da proteína nativa necessários para um adequado desempenho celular. Assim, os principais objetivos desta monografia foram, através de pesquisa bibliográfica e recorrendo a base de dados genéticas, identificar os polimorfismos de nucleótido único que se encontram associados às repetições expandidas, bem como as terapias de silenciamento alelo específicas já estudadas para cada uma das doenças de poliglutaminas.
