dc.description.abstract | Nanopartículas - Um conceito emergente que se tem estendido amplamente pelo mundo, com inúmeras aplicações no campo biomédico, desde intervenções terapêuticas a diagnósticas. Contudo, quão perigosas estas “partículas do tamanho de vírus” poderão ser?Esta é uma questão que a Nanotoxicologia visa responder, particularmente pelo facto de que, devido ao tamanho extremamente pequeno das nanopartículas e pela sua reatividade aumentada, em comparação com as respetivas matérias-primas, estas partículas têm uma probabilidade acrescida de serem internalizadas pelas células e de interagirem com o material genético, com a possibilidade de induzirem danos no ADN.O Ensaio do Cometa, ou Eletroforese em Gel de Célula-Única (SCGE), destaca-se pela sua capacidade em detetar quebras nas cadeias de ADN, em células eucarióticas. Deste modo, este ensaio apresenta um grande potencial na avaliação das interações das nanopartículas com as células, no que diz respeito à genotoxicidade. Assim sendo, aqui apresentamos uma visão geral das características e etapas envolvidas no Ensaio do Cometa, assim como uma revisão e compilação de algumas das nanopartículas que estão mais propensas ao contacto com os sistemas biológicos e que, adicionalmente, possuem o potencial de induzir, direta ou indiretamente, genotoxicidade (ex:. nanopartículas metálicas), através de diversas vias de administração (nanoformulações orais, tópicas, inalatórias, oculares e parenterais).Na era do boom das nanopartículas, onde as guidelines para a sua avaliação ainda são bastante limitadas, é urgente garantir a sua segurança, juntamente com a sua qualidade e eficácia. Para alcançar esse objetivo, acreditamos que o Ensaio do Cometa poderá ser uma ferramenta essencial e uma fonte confiável para se alcançar uma melhor avaliação nanotoxicológica. | por |
dc.description.abstract | Nanoparticles - An emerging concept widely used worldwide, with several applications in the biomedical field, from therapeutics to diagnostics. However, how much hazardous these “virus-sized particles” can be? This is a question that Nanotoxicology aims to answer, particularly when it comes to the fact that, due to the extremely small size of nanoparticles and enhanced reactivity compared to the respective bulk materials, these particles have a high probability of being internalized by cells and interact with the genetic material, with the possibility of inducing DNA damage.The Comet Assay, or Single-Cell Gel Electrophoresis (SCGE), stands out for its capacity of detecting DNA strand breaks in eukaryotic cells. Therefore, it has a huge potential in genotoxicity assessment of nanoparticles and respective cells’ interactions. Here we present an overview of the characteristics and steps involved in the Comet Assay, as well as a review and compilation of some of the nanoparticles that are mostly in contact with biological systems, and additionally have the potential of, direct or indirectly, inducing genotoxicity (e.g.metal nanoparticles), through several routes of administration (oral, skin, inhaled, ocular and parenteral nanoformulations). In a nanoparticles boom era, where guidelines for their evaluation are still very limited, it is urgent to guaranty their safety, along with their quality and efficacy.
