Abstract: A doença de Alzheimer (DA) é a principal causa de demência nos idosos, sendo caraterizada pela perda progressiva de memória. Esta patologia caracteriza-se pela acumulação do péptido beta-amilóide (Aβ), que é considerado um agente causador da disfunção e perda sináptica. Os astrócitos são as células mais abundantes no cérebro, tendo um importante papel na atividade da sinapse e processamento da informação. Estas células alteram a transmissão sináptica através da libertação de gliotransmissores, como ATP e glutamato, e por captarem glutamato e GABA da fenda sináptica. Outra caraterística importante dos astrócitos é a geração de sinais elaborados de Ca2+ e de ondas de Ca2+, que regulam as funções destas células, como a exocitose e a comunicação entre astrócitos através da propagação de sinais de Ca2+. Assim, alterações nas funções dos astrócitos podem contribuir para o processo neurodegenerativo na DA.Os principais objetivos deste estudo foram investigar: i) de que modo é que a exposição ao Aβ1-42 afeta as dinâmicas de Ca2+ e a libertação de gliotransmissores e os mecanismos que medeiam estes eventos e ii) o envolvimento dos recetores A2A de adenosina (A2AR) nas dinâmicas de Ca2+ em astrócitos. Para alcançar estes objetivos foram estabelecidas tarefas para definir: (i) de que modo é que a exposição ao Aβ1-42 afeta a libertação de gliotransmissores por exocitose ou por hemicanais, (ii) se as dinâmicas de Ca2+ são afetadas em condições que mimetizam a DA, e (iii) se os A2AR regulam as dinâmicas de Ca2+.Assim, foram utilizadas culturas primárias de astrócitos incubadas com o péptido sintético Aβ1-42, para mimetizar condições de DA, e células não tratadas (controlo; condições não patológicas). Para avaliar o efeito do Aβ1-42 na libertação de gliotransmissores por exocitose e hemicanais, foram utilizadas ferramentas farmacológicas: BAPTA-AM, EGTA ou bafilomicina A1, que previnem a exocitose, e Gap19 ou duloxetina, que bloqueiam os hemicanais. Os níveis extracelulares de ATP e glutamato foram quantificados utilizando ensaios enzimáticos luminescentes e colorimétricos. As dinâmicas de Ca2+ promovidas pela estimulação com ATP foram avaliadas através de microscopia in vivo utilizando a sonda fluorescente Fluo-4 em astrócitos controlo e expostos a Aβ1-42. Ambos os grupos foram expostos a Xestospongina-C ou EGTA para avaliar a contribuição do efluxo de Ca2+ através dos recetores de inositol trifosfato (IP3R) presentes no reticulo endoplasmático (RE) e do influxo de Ca2+ extracelular no aumento do Ca2+ citosólico, essencial para desencadear as ondas de Ca2+. Com o intuito de investigar se os A2AR afetam as dinâmicas de Ca2+ foram realizadas experiências de microscopia em astrócitos tratados com o antagonista seletivo dos A2AR SCH58261 em condições controlo ou de DA.Os resultados obtidos mostram que a libertação de ATP e glutamato aumentou com a exposição ao Aβ1-42, sendo que o ATP foi libertado principalmente por exocitose (processo dependente de Ca2+) e hemicanais. O bloqueio da exocitose não afetou significativamente a libertação de glutamato, no entanto o Gap19 reduziu (p>0,05) a libertação de glutamato em astrócitos tratados com Aβ1-42, sugerindo que em condições de DA o aumento na libertação de glutamato poderá dever-se a outros mecanismos. Também observámos que as respostas de Ca2+ à estimulação com ATP são afetadas com a exposição ao Aβ1-42, observando-se uma diminuição da amplitude na resposta induzida pelo ATP, no entanto a sua duração era significativamente maior, aumentando a magnitude da resposta. Relativamente à origem do Ca2+ envolvido na resposta ao ATP, em condições não patológicas a principal fonte é o RE, através da ativação dos IP3R, no entanto em condições de DA tanto o Ca2+ proveniente do RE (IP3R) como o Ca2+ extracelular contribuem significativamente para o aumento da resposta. Na presença do antagonista seletivo dos A2AR, SCH58261, a amplitude e magnitude da resposta de Ca2+ induzida por ATP sofreram uma diminuição, sendo as respostas semelhantes no controlo e células expostas ao Aβ1-42. Estes resultados indicam que o bloqueio dos A2AR permite recuperar as alterações induzidas pelo Aβ1-42 nas respostas de Ca2+, principalmente na amplitude e magnitude da resposta ao ATP. Como os A2AR parecem estar envolvidos nas ondas de Ca2+, começámos a definir as condições para silenciar estes recetores em astrócitos em cultura.Em conjunto, estes resultados indicam que condições que mimetizam a DA causaram uma diminuição significativa na libertação de ATP por exocitose e hemicanais e nas dinâmicas de Ca2+ após estimulação. Tendo em conta que a exocitose é um processo dependente de Ca2+, o aumento na libertação de ATP por exocitose pode relacionar-se com a maior magnitude da resposta de Ca2+ em astrócitos expostos ao Aβ1-42. O bloqueio dos A2AR permite recuperar o impacto do Aβ1-42 nas ondas de Ca2+ nos astrócitos. Assim, podemos especular que os A2AR astrocíticos podem ser um importante alvo terapêutico em DA.
Alzheimer’s disease (AD) is the main cause of dementia in elderly and is characterized by a progressive loss of memory. The accumulation of amyloid-beta peptides (Aβ) is one hallmark of AD and is believed to cause synaptic dysfunction and loss. Astrocytes, the most abundant cells in the brain, are important participants in the synapse function and information processing in brain. These glial cells shape the synaptic function through the release of gliotransmitters, like ATP and glutamate, and by performing the uptake of glutamate and GABA from the synaptic cleft. Another important characteristic of astrocytes is formation of elaborated Ca2+ signals and the generation of Ca2+ waves that are involved in several mechanisms like exocytosis and communication between astrocytes through the propagation of Ca2+ signalling. Thus, alterations in astrocytes functions might contribute to neurodegenerative process in AD.The major aims of this study were to investigate: i) how the Aβ1-42 peptides affect astrocytic Ca2+ dynamics and gliotransmitter release and also the mechanisms underlying these events and ii) the involvement of adenosine A2A receptors (A2AR) in astrocytic Ca2+ dynamics. To achieve these aims, we established three tasks to define: (i) how Aβ1-42 exposure affect astrocytic ATP and glutamate release through exocytosis and hemichannels, (ii) if astrocytic Ca2+ dynamics are affected in AD-like conditions, and (iii) whether A2AR regulate Ca2+ dynamics.To accomplish our aims, we used primary cultures of astrocytes incubated with the synthetic peptide Aβ1-42 to mimic AD-like conditions or non-treated (control) cells, which correspond to non-pathologic conditions. To evaluate the effect of Aβ1-42 on gliotransmitter release mediated by exocytosis and hemichannels, we used pharmacological tools: BAPTA-AM, EGTA or bafilomycin A1 to prevent exocytosis and Gap19 or duloxetine to block hemichannels. The extracellular levels of ATP and glutamate were quantified by luminescent and colorimetric enzymatic assays. Ca2+ dynamics triggered by ATP stimulation was performed by live single-cell Ca2+ measurements, using a fluorescent probe Fluo-4, in astrocytes exposed to Aβ1-42 and in control astrocytes. In both conditions, cells were exposed to Xestospongin-C (Xe-C) or EGTA to evaluate the contribution of Ca2+ efflux through inositol triphosphate receptors (IP3R) from endoplasmic reticulum (ER) or of extracellular Ca2+ influx in cytosolic Ca2+ increase, essential for triggering Ca2+ waves. To investigate whether A2AR affect Ca2+ dynamics, cell-live Ca2+ imaging in astrocytes challenged with ATP was performed in the presence of the selective antagonist of A2AR SCH58261 in both control and AD-like conditions.Our data showed that Aβ1-42-exposure increased astrocytic ATP and glutamate release, being the ATP released mainly through Ca2+-dependent exocytosis and hemichannels. However, the blockade of exocytosis did not significantly affect glutamate release, whereas Gap19 caused a reduction (P>0.05) in the release of this gliotransmitter in Aβ1-42-treated astrocytes, suggesting that under AD-like conditions the increased glutamate release might be related with other mechanisms. Moreover, we observed that the astrocytic Ca2+ response to ATP stimulation was affected by Aβ1-42 (compared with control cells), being detected a decrease in amplitude (peak) of the response, though its duration was significantly increased and, consequently, the magnitude of Ca2+ response was higher in AD conditions. Regarding Ca2+ sources involved in astrocytic responses to ATP, in non-pathologic conditions the main Ca2+ source was the ER through IP3R activation, whereas in AD-like conditions both IP3R-mediated Ca2+ release from ER and extracellular Ca2+ influx contributed significantly to the augmented response. In the presence of A2AR selective antagonist, SCH58261, the amplitude and the magnitude of ATP-evoked Ca2+ response were reduced, being these responses similar in control and Aβ1-42-treated cells. These data suggest that the blockade of A2AR was able to recover the alterations induced by Aβ1-42 in astrocytic Ca2+ responses, mainly in amplitude and magnitude, evoked by ATP stimulation. Since the A2AR seemed to be involved in astrocytic Ca2+ waves, we started setting the conditions to knockdown these receptors in cultured astrocytes.Taken together, the results indicated that AD-like conditions significantly increased the astrocytic ATP release, which occurred through exocytosis and HC, and the magnitude of Ca2+ response to ATP stimulation. Since exocytosis is a Ca2+-dependent mechanism, the increased exocytotic ATP release might be related with the augmented Ca2+ responses observed in astrocytes exposed to Aβ1-42. The blockade of A2AR was able to recover the changes in Ca2+ waves in astrocytes exposed to Aβ1-42. Thus, we can speculate that astrocytic A2AR are a valuable therapeutic target for AD.
