Abstract: A Diapausa embrionária, em que o desenvolvimento embrionário ou acontece lentamente ou é temporariamente suspenso, constitui um mecanismo sofisticado para garantir que o desenvolvimento ocorra em condições adequadas, otimizando e salvaguardando a sobrevivência da descendência. Este mecanismo é, também, caracterizado por uma diminuição da biossíntese, em que o embrião permanece num estado hipometabólico, e aumenta a resistência ao stress. Recentemente, foi descoberto que o mammalian target of rapamycin (mTOR) tem um papel fulcral a regular a diapausa, pois a sua inibição farmacológica, induz um estado semelhante à diapausa, tanto in vivo quanto in vitro, mantendo o embrião em fase de blastocisto durante semanas. Esta inibição provou-se ser reversível e mais importante, estendida a células estaminais pluripotentes, induzindo um novo estado de pluripotência em pausa. A via mTOR é uma importante via integrativa que coordena vários processos celulares em resposta a diversos sinais, entre os quais a disponibilidade de aminoácidos, especialmente leucina e arginina. Aliás, a ausência destes dois aminoácidos causa a interrupção do desenvolvimento embrionário in vitro através de efeitos no mTOR, embora estudos anteriores se concentrassem apenas em efeitos no trofoblasto. Assim, o principal objetivo deste estudo foi induzir este novo estado de pluripotência pausada através da remoção de Leucina e Arginina, evitando, desta forma, abordagens farmacológicas. Os nossos resultados sugerem que a ausência de ambos os aminoácidos foi capaz de reduzir a proliferação celular, afetando a progressão do ciclo celular o que é refletido por um aumento de células na fase G1, acompanhado também por uma redução no metabolismo glicolítico a níveis basais. Os resultados mostram também que a pluripotência não foi afetada e todos os efeitos observados da remoção de aminoácidos eram reversíveis. Juntamente com os resultados da inibição farmacológica do mTOR, os nossos resultados sugerem que a remodelação metabólica é a principal potencializadora da indução do estado de pluripotência pausada.
Embryonic Diapause, in which embryo development is very slow or temporarily arrested, constitutes a very sophisticated mechanism to ensure that proper conditions are available for development to occur, safeguarding and optimizing offspring survival. This mechanism is also characterized by a decrease in biosynthesis, hypometabolic state and improvement on stress resistance. Recently, the mammalian target of rapamycin (mTOR) was found to be a central player regulating diapause, since its inhibition, using a pharmacological approach, induces a diapause-like state, either in vivo or in vitro, maintaining the embryo at the blastocyst stage for weeks. This inhibition was proven to be reversible and more importantly, extended to pluripotent stem cells, inducing a novel state of paused pluripotency. The mTOR pathway is a major integrative pathway that coordinates several cellular processes in response to a variety of environmental cues, including amino acid availability, especially Leucine and Arginine. Moreover, absence of these two amino acids was previous shown to arrest embryo development in vitro in an mTOR dependent-fashion, though studies only focused on trophoblast outgrow. Therefore, the main goal of this study was to induce this novel paused pluripotent state by Leucine and Arginine withdrawal from the culture medium, to avoid pharmacological approaches. Our results suggest that absence of both amino acids reduced cell proliferation by affecting cell cycle progression, reflected by an increase of cells in the G1 phase accompanied by a reduction in glycolytic metabolism at basal levels. Moreover, pluripotency was not affected and all observed effects of aminoacid withdrawal were reversible. Our results, together with mTOR inhibitor results, suggest that metabolic remodeling is the main mediator of the induction of pluripotency paused state.
