da história da humanidade pelos seus efeitos terapêuticos e curativos. Estas plantas
conseguem sintetizar uma grande variedade de fitoquímicos biologicamente ativos, dos quais
se podem destacar os compostos fenólicos. O interesse no estudo destes metabolitos tem
aumentado exponencialmente nestes últimos anos, uma vez que se pensa que estejam
intimamente associados a diversas propriedades benéficas para a saúde, entre as quais a
capacidade antioxidante e anti-inflamatória.
Geranium robertianum é uma espécie de planta que tem sido muito utilizada na
medicina tradicional devido as suas propriedades benéficas. Esta planta é vulgarmente
utilizada na forma de infusões ou pastas, atuando como agente anti-inflamatório,
antioxidante, anti-hepatotóxico, diurético e antidiabético. Com base nestes alegados
benefícios, existem atualmente no mercado diversos suplementos alimentares à base de
extratos de G. robertianum L.
Neste trabalho pretendeu-se caracterizar os extratos etanólicos de G. robertianum,
tendo-se para tal recorrido ao seu fracionamento sequencial com diferentes solventes
orgânicos. Este fracionamento tem como objetivo a obtenção de frações enriquecidas em
diversos compostos, por forma a isolar e caraterizar compostos através da espectroscopia
de ressonância magnética nuclear. Por outro lado, o conhecimento dos extratos etanólicos
permite também conhecer melhor a composição fitoquímica da planta, percebendo se existe
potencial para que esta possa vir a ser explorada com fins terapêuticos ou como
formulações para nutracêuticos.
Métodos: A composição fitoquímica dos extratos etanólicos foi conseguida pela
análise destas e/ou de frações obtidas por solubilização sequencial a quente com n-hexano,
diclorometano, acetato de etilo, acetona e metanol (frações EtOH1_Hex e EtOH2_Hex,
EtOH1_Dic e EthOH2_Dic, respetivamente). A caraterização dos componentes voláteis e
fenólicos dos extratos e frações foi efetuada com recurso à análise de GC-MS e/ou UHPLCDAD-
ESI/MSn, tendo-se ainda procedido ao isolamento por HPLC de dois compostos e
efetuado análise de 1H RMN, com a finalidade de elucidar a sua estrutura. Adicionalmente, as
atividades antioxidante e anti-inflamatória dos extratos etanólicos e respetivas frações foram
avaliadas pelo método de Folin-Ciocalteu e de captação de NO•, respetivamente.
Resultados: Os extratos etanólicos de G. robertianum analisados em GC-MS,
mostraram na sua composição, álcoois, monossacarídeos, ácidos carboxílicos e ácidos

gordos, sendo os ácidos gordos palmítico e linoleíco os elementos predominantes nas
análises efetuadas. Por sua vez, a análise dos extratos etanólicos e respetivas frações
efetuada por UHPLC-DAD-ESI/MSn permitiu concluir que os compostos fenólicos
predominantes correspondem à hidroxidifenilenogaloil-hexosídeo (corilagina), brevifolina,
ácido elágico e ao composto desconhecido [M-H] ¯= m/z 319. O ácido galoil-quínico , ácido
dicafeoil-quínico e a flavona luteolina também foram identificados, embora com menor
intensidade. Adicionalmente, a análise de 1H RMN dos compostos, hidroxidifenilenogaloilhexosídeo
e o composto com [M-H]¯= m/z 319 permtiu identificar alguns dos seus sinais
característicos, muito embora não tenha permitido elucidar toda a estrutura destes
compostos. No que diz respeito ao composto [M-H]¯= m/z 319, foi possível observar a
presença de sinais na zona dos aromáticos assim como na zona alifática do espetro.
Relativamente à atividade antioxidante dos extratos etanólicos e frações, EtOH2
mostrou melhor resultado (190 μg±3 GAE/mg extrato), seguindo-se das frações de acetato
de etilo e acetona (EtOH2_Acett, EtOH1_Acetn e EtOH2_Acetn, com 148±6 μg GAE/mg
extrato, 178±1 μg GAE/mg extrato e 170±0,7 μg GAE/mg extrato, respetivamente).
Relativamente à capacidade de captação de NO•, a amostra que mostrou um menor IC50 foi
EtOH2_Acett com 0,09±0,02 mg/mL, seguindo-se as amostras de EtOH1 e EtOH2 com um
IC50 de 0,14±0,01 mg/mL e 0,14±0,03 mg/mL, respetivamente.
Conclusões: Este estudo contribuiu para o conhecimento fitoquímico dos extratos
etanólicos de G. robertianum. Relativamente às atividades biológicas, os extratos etanólicos
mostraram resultados promissores para atividades antioxidantes, além disso também
revelaram ser eficazes na captação de NO•.
A vasta gama de compostos identificados em cada extrato analisado, assim como as
proeminentes atividades biológicas, são claramente um contributo para a valorização de G.
robertianum. Introduction: Medicinal plants are recognized and widely used through history for their therapeutic and healing benefits. Those plants have the capability of synthesizing a great variety of biologically active phytochemicals, among them the phenolic compounds. The study of these metabolites has increased exponentially in recent years, since they are claimed to be intimately associated with several beneficial properties to health, highlighting the antioxidant and anti-inflammatory capability. Geranium robertianum is a plant widely used in traditional medicine due to its beneficial properties. This plant is commonly used as infusion or paste, acting as antiinflammatory agent, antioxidant, anti-hepatotoxic, diuretic and antidiabetic. Based on these alleged benefits, there are currently several dietary supplements on the market based on G. robertianum L extracts . This study aimed to characterize the ethanolic extracts of G. robertianum. For that, sequential fractionation of ethanolic extracts were performed with different organic solvents. This approach intended to allow the obtaining of fractions enriched in distinct compounds, enabling the isolation and their further structural characterization by nuclear magnetic resonance spectroscopy. On the other hand, the knowledge of the ethanolic extracts of a plant leads to the understanding of its phytochemical composition, thus facilitating the understanding of their potential exploitation for usage in therapeutics or nutraceuticals formulations. Methods: The phytochemical composition of ethanol extracts was achieved by their analysis and/or of their respective fractions obtained by hot solubilization with n-hexane, dichloromethane, ethyl acetate and methanol (fractions EtOH1_Hex and EtOH2_Hex, EtOH1_Dic and EthOH2_Dic, EtOH1 respectively). The characterization of volatile and phenolic components of the extracts and fractions was performed using the GC-MS and/or UHPLC-DAD-ESI/MSn analysis. In addition, two phenolic compound were isolated by HPLC and their 1H RMN analysis was performed in order to achieve structural details. Moreover, the antioxidant and anti-inflammatory potential of ethanolic extracts and fractions were evaluated respectively by the Folin-Ciocalteu and NO• scavenging assays. Results: G. robertianum ethanol extracts analyzed by GC-MS presented in its composition alcohols, monosaccharides, carboxylic and fatty acids, being the fatty acids palmitic and linoleic the prevalent elements on the analysis performed. On the hand, the the UHPLC-DAD-ESI/MSn analysis of ethanolic extracts and their fractions allowed to conclude that the main phenolic compounds correspond to galloyl-HHDP-hexoside (corilagin) IV brevifolin, ellagic acid and an unknown compound [MH]¯at m/z 319. Galloyl-quinic acid, dicaffeoyl-quinic acid and flavone luteolin were also identified, though they showed less intensity. In addition, the 1H NMR analysis of galloyl-HHDP-hexoside and [M-H] ¯= m/z 319 allowed to identify some of their typical 1H signals, althought the complete structural elucidation of these compounds was not possible to achive. Relatively to compound [MH] ¯at m/z 319 it was possible observed the presence of signals in aromatic and aliphatic region. Regarding the antioxidant activity of ethanol extracts and fractions, EtOH2 (190 ± 3 μg GAE/mg extract) showed the best results, followed the ethyl acetate and acetone fractions (EtOH2_Acett, EtOH1_Acetn and EtOH2_Acetn at 148 ± 6 μg GAE/mg extract, 178 ± 1 μg GAE/mg extract and 170 ± 0.7 μg GAE/mg extract, respectively). For the NO• scavenging assay, the sample EtOH2_Acett showed the lowest IC50 (0.09 ± 0.02 mg /mL), followed by EtOH1 and EtOH2 samples, with IC50 respectively 0.14 ± 0.01 mg/mL and 0.14 ± 0.03 mg/mL. Conclusions: This study contributed to the phytochemical knowledge of the ethanol extracts of G. robertianum. Concerning the biological activity, the ethanol extracts showed promising results for antioxidant activity and proved to be effective in NO• scavenging. Hence, overall, the wide range of compounds identified with each extract, as well as the prominent biological activities, are clearly a contribution to the valorization of G. robertianum. |
