Abstract: Resumo A amêijoa asiática, Corbicula fluminea (Müller, 1774) é um molusco bivalve de água doce e é uma das espécies invasoras de ecossistemas aquáticos mais difundida no mundo. Para além dos efeitos nefastos nos ecossistemas, esta peste é conhecida pela sua acção bioincrustante nas indústrias hidrodependentes. A introdução desta espécie em Portugal data de 1980, no entanto, existe um largo desconhecimento acerca da peste por parte das indústrias nacionais e a monitorização das fontes de água primárias será crucial para a minimização da sua dispersão e impactes. Esta tese teve dois objectivos. Primeiro, sistematizar informação acerca da monitorização da amêijoa asiática para fins de controlo em ambiente industrial. Depois, contribuir para a criação de soluções de controlo inovadoras através da avaliação e interpretação da toxicidade de misturas de potenciais agentes de controlo químico. No âmbito da primeira componente do trabalho, foi elaborado um texto que pretende funcionar como um auxiliar paras as indústrias nacionais, dando a conhecer as características de C. fluminea e ajudando no projecto de um sistema de monitorização de C. fluminea. No âmbito da segunda componente deste trabalho, foi testado o potencial de três combinações binárias de poli(cloreto de dimetildialilamónio) (polyDADMAC) com cloreto de potássio, niclosamida e dimetoato para o controlo da amêijoa asiática, realizando testes de toxicidade à escala laboratorial. O polyDADMAC foi mantido como elemento comum nas misturas de forma a explorar diferentes tipos de interacções e avaliar até que ponto o desempenho do tratamento combinado pode ser melhorado manipulando o modo de acção dos químicos individuais. Destes estudos concluiu-se que a niclosamida e o dimetoato são tóxicos para a amêijoa asiática na gama de concentrações testadas, podendo eventualmente equacionar-se a sua aplicação individual em programas de controlo. No geral, o efeito conjunto dos compostos nas misturas testadas é antagonista e, por isto, não são promissoras para o controlo da amêijoa asiática em ambiente industrial. Com o objectivo de auxiliar a compreensão do carácter antagonista dos químicos foi efectuado um estudo preliminar do modo de actuação do polyDADMAC e do dimetoato em C. fluminea bem como da sua potencial interacção. Este estudo indica que o polyDADMAC não afecta nem a capacidade de osmorregulação das células nem interage com a actividade de enzima acetilcolinesterase da amêijoa asiática. Este estudo confirma ainda que o dimetoato inibe a actividade da enzima acetilcolinesterase.
Abstract The freshwater bivalve Corbicula fluminea (Müller, 1774), commonly known as the Asian clam, is one of the most invasive species of aquatic systems around the world. In addition to the adverse effects on ecosystems, this pest is well-known for its biofouling action on freshwater-dependent industries. While the introduction of this species in Portugal dates back to 1980, there is still a lack of knowledge about the species amongst national industries and the monitoring of primary water sources will be crucial to minimize the pest dispersion and impacts. This thesis has two main objectives objectives: (i) systematize information about the Asian clam monitoring for control purposes in industrial environments; (ii) contribute to the development of innovative control solutions through the evaluation and interpretation of the toxicity of mixtures of chemical control agents. Regarding the first component of the study, a text was prepared to assist national companies to project monitoring systems for C. fluminea. The second component of this study was to test the potential of three binary mixtures of poly(diallyldimethyl-ammonium chloride) (polyDADMAC) combined with potassium chloride, niclosamide and dimethoate to control the Asian clam, by performing toxicity tests at the laboratory scale. PolyDADMAC was kept as a common ingredient in the three mixtures in order to explore different types of interactions and assess the extent to which the performance of the combined treatment can be improved by manipulating the mode of action of individual chemicals. These studies showed that niclosamide and dimethoate are toxic to the Asian clam, and their individual application in control programs can be considered. The results also showed that, in general, the combined effect of the compounds is antagonist and, therefore, the mixtures are not promising for control of Asian clams in industrial environment. In order to understand the antagonist nature of the chemicals, a preliminary study was conducted to elucidate the modes of action of polyDADMAC and dimethoate and their interactions in C. fluminea. This study indicates the polyDADMAC neither affects the ability of osmotic regulation of the cells nor interacts with the activity of the enzyme acetylcholinesterase. This study also indicates that dimethoate inhibits the activity of the enzyme acetylcholinesterase.
