vários tecidos, no entanto, também são responsáveis pelo crescimento de certos tumores,
entre os quais, o da mama. Neste tipo de cancro da mama os tratamentos hormonais são os
mais adequados. Os estudos mais recentes demonstraram um papel de destaque dos
inibidores da aromatase, principalmente os de terceira geração. A aromatase está presente
em vários tecidos e é responsável pela biossíntese de estrogénios, principalmente nas
mulheres pós menopausa em que a função ovárica cessa e a produção de estrogénios ocorre
através da aromatase nos tecidos periféricos. A inibição da aromatase é então uma boa
estratégia terapêutica. Os inibidores da aromatase podem ser esteróides ou não esteróides
e interagem com o local activo da enzima. Actualmente já existem inibidores da aromatase
de terceira geração, sendo que estes possuem maior eficácia e melhor perfil de toxicidade
que o tamoxifeno, que era o tratamento padrão na maioria das doentes em pós menopausa.
No entanto, também apresentam efeitos secundários e episódios de resistência o que
justifica a pesquisa de novos IA mais eficazes e selectivos. Estrogens are required for regular growth and development of many tissues, however, they are also responsible for the growth of certain cancers, including the breast cancer. In this type of breast cancer hormonal treatments are most suitable. The most recent studies have demonstrated a prominent role of aromatase inhibitors, especially the third generation ones. Aromatase is present in various tissues and is responsible for the biosynthesis of estrogens, particularly in postmenopausal women in whom ovarian function ceases and estrogen production occurs through aromatase in peripheral tissues. Therefore, inhibition of aromatase is a good therapeutic strategy. Aromatase inhibitors can be steroids or non-steroids and interact with the active site of the enzyme. Currently there are already third generation aromatase inhibitors, and these have higher efficacy and better toxicity profile than tamoxifen , which was the standard treatment for most patients in postmenopausal. However, they also have side effects and episodes of resistance which justifies the search for more effective and selective new aromatase inhibitors. |
