em duas ou mais formas cristalinas, devido aos diferentes arranjos das moléculas no cristal. É
um fenómeno que pode causar alterações na qualidade durante a produção ou
armazenamento de medicamentos, podendo influenciar a sua eficácia. Assim, é essencial
compreender o comportamento de estado sólido do fármaco e selecionar criteriosamente a
forma cristalina ideal para desenvolvimento. Diversas técnicas analíticas são usadas para a
identificação e caracterização das diferentes formas polimórficas.
Os co-cristais farmacêuticos representam uma classe de sólidos cristalinos que oferece
alternativas para otimizar as propriedades físico-químicas de ativos pouco solúveis em água.
A sua formação depende de grupos funcionais complementares entre um ativo farmacêutico
e uma molécula biologicamente segura, denominada co-formador. Hoje em dia, o valor dos
co-cristais estende-se ao domínio da propriedade intelectual, representando uma boa
oportunidade para patentear. Polymorphism is defined as the ability of a compound to exist in two or more crystalline forms due to different arrangements of the molecules in the crystal. It is a phenomenon which can cause quality deviation during manufacture or storage of medicines and may influence its effectiveness. Thus, it is essential to understand the solid-state behavior of the drug and carefully select the optimal solid form to development. Various analytical techniques are used on the identification and characterization of the different polymorphic forms. Pharmaceutical co-crystals are a class of crystalline solids offering the prospect of better alternatives to optimize physical-chemical properties of poorly-soluble drugs. Their formation relies on complementary functional groups between the drug and a biologically safe molecule, named co-former. Nowadays, the value of co-crystallization extends to the field of intellectual property, representing a good opportunity to patent. |
