dc.description.abstract | O cancro continua a ser uma das doenças com maior impacto na sociedade, não só pela gravidade como também pela frequência, sendo detetados anualmente números crescentes de novas pessoas afetadas. Apesar dos avanços no tratamento oncológico, a quimioterapia continua a fazer parte da terapêutica convencional. No entanto, grande parte dos anticancerígenos usados acarreta problemas como: solubilidade, toxicidade e pouca especificidade, comprometendo a eficácia do tratamento e podendo induzir resistência. As nanopartículas lipídicas (NL) surgem assim como um sistema de transporte de anticancerígenos muito promissor. Esta dissertação aborda a aplicação das várias gerações de nanopartículas lipídicas no transporte de anticancerígenos, em particular na resolução dos problemas a estes associados. As perspetivas futuras aliam as vantagens das NL à co-encapsulação de anticancerígenos com quimiossenssibilizadores e ao targeting de forma a tornar a terapêutica mais eficaz, especialmente quando existem mecanismos de resistência. | pt |
dc.description.abstract | Cancer is one of the most devastating diseases affecting the life of many people around the world, with an increased number of new diagnosed cases every year. Despite the advances in oncology treatment, chemotherapy still remains the standard of care. However, a large part of the conventionally used anticancer drugs has problems related to solubility, low toxicity and specificity, thus compromising the effectiveness of the treatment, also being able to induce resistance. Lipid nanoparticles (LN) are a very promising anticancer drug transport system. This dissertation addresses the application of the several generations of lipid nanoparticles in the transport of anticancer drugs, giving particular insight to the problems they are able to solve. A future prospective about the use of LN for the co-encapsulation of anticancer and chemosensitizer compounds, and their targeting ability, in order to provide a more effective treatment, especially in the case of resistance mechanisms, is finally presented. | pt |
