O sistema CRISPR/Cas9 (clustered regularly short palindromic repeats-CRISPR associated nucleases) tem-se destacado devido à sua versatilidade e simplicidade de manuseamento em laboratório. O seu potencial terapêutico tem aumentado com o conhecimento cada vez mais pormenorizado dos seus mecanismos de atuação e das diferentes estratégias de modificação usadas para melhorar a especificidade e eficiência.
Neste trabalho são apresentados os mais recentes avanços na tecnologia CRISPR/Cas9 que a aproximam da sua aplicação na resolução de doenças genéticas e epigenéticas. Increasing advances in gene therapy have revolutionized the way we think about everything around us, particularly in healthcare. The CRISPR/Cas9 system (Clustered Regularly Short Palindromic Repeats-CRISPR associated nuclease 9) has been highlighted due to its versatility and simplicity to be handed in the laboratory. The therapeutic potential of this gene editing technology has increased with the progressively detailed knowledge of its mechanisms of action and different strategies used in order to raise its specificity and efficiency. This dissertation presents the latest advances in CRISPR/Cas9 technology, which are suggestive of its application as a tool for the resolution of genetic and epigenetic diseases. |
