dc.description.abstractThe neurotrophin BDNF regulates the maturation of the postsynaptic density of hippocampal synapses and is a key modulator of activity-dependent modifications of synaptic strength in the Central Nervous System. Physiological and biochemical evidences implicate the ionotropic glutamate receptors as targets for BDNF modulation. In this study we show that acute stimulation of hippocampal neurons with BDNF differentially upregulate the protein levels of GluR1, GluR2 and GluR3 AMPA receptor subunits, and NR1, NR2A and NR2B NMDA receptor subunits, by a mechanism sensitive to transcription and translation inhibitors. Accordingly, BDNF increase the mRNA levels for GluR1, GluR2, NR1, NR2A and NR2B subunits. Biotinylation studies showed that stimulation with BDNF selectively increased the amount of GluR1 associated with the plasma membrane, by a mechanism sensitive to emetine, and enhanced the surface levels of NR1, NR2A and NR2B, although with different kinetics. The rapid upregulation of plasma membrane-associated NMDA receptor subunits was correlated with an increase in the intracellular calcium response to NMDA, presumably through an enhancement of the NMDA receptor activity. BDNF also induced GluR1 phosphorylation on Ser831 through activation of PKC and CaMKII. Chelation of endogenous extracellular BDNF with TrkB-IgG selectively decreased GluR1 protein levels in 14 DIV cultures of hippocampal neurons, but had no effect of NMDA receptors. Moreover, BDNF promoted synaptic delivery of homomeric GluR1 AMPA receptors in cultured organotypic slices, by a mechanism independent of NMDA receptor activation. Taken together, the results indicate that BDNF increases the abundance of AMPA and NMDA receptor subunits in hippocampal neurons, and induces their delivery to the plasma membrane thereby increasing the receptor activity in hippocampal neurons.-
dc.description.abstractA neurotrofina BDNF regula a maturação da densidade pós-sináptica nos neurónios do hipocampo e tem um papel fundamental na modulação da actividade das sinapses do Sistema Nervoso Central. Estudos fisiológicos e bioquímicos revelaram que os receptores ionotrópicos do glutamato são regulados pelo BDNF. Neste estudo, verificámos que a estimulação de neurónios do hipocampo em cultura com BDNF aumenta a abundância das subunidades GluR1, GluR2 e GluR3 dos receptores AMPA, e das subunidades NR1, NR2A e NR2B dos receptores NMDA, através de um mecanismo dependente da tradução e transcrição. De igual modo, o BDNF aumentou os níveis de RNAm das subunidades GluR1, GluR2, NR1, NR2A e NR2B. Estudos de biotinilação revelaram que a estimulação com BDNF também aumentou a quantidade de GluR1 na membrana plasmática, por um mecanismo sensível ao inibidor da tradução emetina. O BDNF também aumentou os níveis de NR1, NR2A e NR2B na membrana, embora com diferentes cinéticas. Ao aumento rápido da quantidade dos receptores NMDA na membrana plasmática correspondeu um incremento na alteração da concentração intracelular de cálcio induzida por NMDA. O BDNF também induziu a fosforilação da subunidade GluR1 na Ser831 através da activação da PKC e da CaMKII. A sequestração do BDNF endógeno presente no meio extracelular com TrkB-IgG diminuiu os níveis totais da subunidade GluR1 em culturas de hipocampo com 14 dias, mas não teve qualquer efeito na abundância dos receptores NMDA. Além disso, o BDNF induziu o direccionamento dos receptores AMPA constituídos apenas pela subunidade GluR1 para a sinapse em fatias organotípicas de hipocampo, através de um mecanismo independente da activação dos receptores NMDA. Os resultados obtidos indicam que o BDNF aumenta a abundância das subunidades dos receptores AMPA e NMDA em neurónios de hipocampo em cultura e induz o seu direccionamento para a membrana plasmática, potenciando a actividade dos receptores.-
