Abstract: O trabalho apresentado nesta dissertação centra-se no estudo de dois processos reacionais distintos: as reações de tautomerismo prototrópico induzidas por irradiação UV e as reações de isomerização conformacional induzidas por irradiação IV. O objetivo do trabalho é clarificar as mecanísticas envolvidas nos processos de fototautomerismo, e discutir os fatores que controlam a isomerização conformacional. Estes fenómenos foram estudados no caso de compostos heteroaromáticos isolados em matrizes criogénicas, nomeadamente o benzimidazol (BzIm), e os três isómeros 5-metoxi-benzimidazol (5M-BzIm), 5-metoxi-6-azaindol (5M-6AzI) e 5-metoxi-4-azaindol (5M-4AzI).É conhecido pelo estudo da literatura que as moléculas análogas ao BzIm, 5M-BzIm, 5M-6AzI e 5M-4AzI, quando expostas à radiação UV, sofrem a quebra da ligação NH acompanhada de formação de radicais, que podem posteriormente recombinar e originar novas formas tautoméricas. Estes radicais, assim como os tautómeros de alta energia, são tipicamente muito reativos. No entanto, nas condições da técnica de isolamento em matrizes criogénicas podem ser estabilizados. Esta técnica de amostragem foi a técnica maioritariamente utilizada no presente estudo. Uma vez estabilizados na matriz, as moléculas podem ser caracterizadas por espectroscopia de infravermelho.O trabalho iniciou-se pela caracterização das propriedades luminescentes dos compostos estudados em solução, observando-se que todos eles apresentam emissão de fluorescência e fosforescência. Seguiu-se o estudo da fotoquímica do BzIm isolado em árgon. Este composto existe inicialmente somente na forma 1H-BzIm, mas isomeriza para dois tautómeros de alta energia, o 5H-BzIm e o 7H-BzIm, quando irradiado no UV. Estes tautómeros novos convertem-se entre si, ou isomerizam de volta ao 1H-BzIm, consoante a irradiação UV utilizada. O mecanismo que rege esta reatividade foi teoricamente analisado e propõe-se que a espécie-chave neste mecanismo deverá ser o radical benzimidazolyl. Os isotopólogos deuterados do BzIm foram preparados na presença de metanol monodeuterado (T= 80 °C). Estes isotopólogos foram posteriormente caracterizados por espectroscopias de RMN e de IV. Observou-se que as posições 1 e 2 do anel de benzimidazol sofrem substituição isotópica, formando-se três isotopólogos deuterados distintos. O estudo da fotoquímica na matriz de árgon revelou que as reações de transferência de hidrogénio/deutério são o principal canal fotoreativo do benzimidazol.Avaliaram-se também os isomerismos tautoméricos do 5M-6AzI e do 5M-4AzI isolados em matrizes de Ar e Xe. Ambos os derivados existem inicialmente na matriz numa única forma tautomérica (1H). Observou-se que o 5M-6AzI isomeriza sob irradiação UV para os tautómeros 3H-5M-6AzI e 6H-5M-6AzI. A formação de 6H-5M-6AzI só foi observada na matriz de árgon, e por intermédio do radical 5M-6AzI-yl. O 5M-4AzI também isomeriza para o tautómero 3H-5M-4AzI. No entanto, não se observou a formação do radical 5M-4AzI-yl ou do tautómero 4H-5M-4AzI. A diferente dependência com o material hospedeiro evidenciou que as reações de tautomerismo observadas deverão ser regidas por dois mecanismos distintos, um de natureza radicalar e dependente do material hospedeiro usado, e outro não.A colocação de um grupo metoxilo na posição 5 do anel de benzimidazol permitiu que os tautómeros 1H-5M-BzIm e 3H-5M-BzIm fossem distinguíveis. Cada um deles pode apresentar duas conformações distintas (syn e anti) conforme a orientação do grupo metoxilo em relação ao anel, tendo-se registado quatro mínimos na superfície de energia potencial do 5M-BzIm, todos eles de energias próximas. A estrutura cristalina do 5M-BzIm foi resolvida por difração de raios X, verificando-se que somente o tautómero 3H-5M-BzIm a constituía, e numa única conformação (anti). No entanto, na fase gasosa e nas matrizes criogénicas, as quatro espécies mais estáveis desta molécula coexistem, com populações próximas das teoricamente previstas. As assinaturas individuais de cada espécie presente na matriz de xénon foram atribuídas espectroscopicamente. Para cada tautómero verificou-se que as populações conformacionais podem ser manipuladas por excitação vibracional monocromática, pela exposição à fonte de luz do espectrofotómetro, ou termicamente.Para o único confórmero do 5M-4AzI isolado em Xe a 10 K, observou-se a sua isomerização conformacional quando exposto à radiação IV monocromática. Para além disso, observou-se que o novo confórmero de alta energia relaxa totalmente, quando sujeito à luz da fonte do espectrofotómetro, ou termicamente (T= 39 K). A racionalização dos resultados observados foi suportada por cálculos teóricos aos níveis B97-1/Def2-TZVP e MP2/Def2-TZVP. O estudo detalhado das SEP do 5M-4AzI e do 5M BzIm permitiu uma análise comparativa e explicou a maior dificuldade de produção e estabilização do confórmero de maior energia no caso do 5M-4AzI, em comparação com os confórmeros de alta energia do 5M-BzIm.
The research presented in this thesis focuses on the study of two distinct reactive processes: the tautomeric reactions induced by UV irradiation and the conformational isomerization reactions induced by IR irradiation. The goal of this work is to clarify the mechanisms involved in the phototautomeric processes, and to discuss the factors that control conformational isomerization. These phenomena were studied for heteroaromatic compounds isolated in cryogenic matrices, namely benzimidazole (BzIm), and the three isomers 5 methoxy-benzimidazole (5M-BzIm), 5-methoxy-6-azaindole (5M-6AzI) and 5 methoxy-4 azaindole (5M-4AzI).It is known from the bibliography that the molecules analogous of BzIm, 5M-BzIm, 5M 6AzI and 5M-4AzI, when exposed to UV-irradiation, undergo the cleavage of NH bond accompanied by production of radicals, which can later recombine and originate new tautomeric forms. Such radicals and high energy tautomers are typically very reactive. However, in the conditions of matrix isolation technique, they can be stabilized. This sampling technique was the main technique used in the present study. Once stabilized in the matrix, these can be characterized using infrared spectroscopy. The research began by characterizing the luminescent properties of the studied compounds in solution. All compounds reveal fluorescence and phosphorescence emission. This was followed by the photochemical study of BzIm isolated in argon. This compound initially exists only in the 1H-BzIm form, but isomerizes to two high energy tautomers, 5H BzIm and 7H-BzIm, when exposed to UV radiation. These new tautomers interconvert, or isomerize back to the 1H-BzIm tautomer, according to the chosen wavelength of UV irradiation. The mechanism which governs this reactivity was theoretically analyzed and it is proposed that the key species in this mechanism should be the benzimidazolyl radical.The BzIm deuterium isotopologues were prepared in the presence of monodeuterated methanol (T= 80 °C). These isotopologues were subsequently characterized by NMR and IR spectroscopies. It was observed that positions 1 and 2 of the benzimidazole ring undergo isotopic substitution, yielding three distinct deuterated isotopologues. The study of photochemistry in an argon matrix revealed that the hydrogen/deuterium transfer reactions are the main photoreaction channel of benzimidazole. The tautomeric isomerizations of 5M-6AzI and 5M-4AzI isolated in Ar and Xe matrices were also studied. Both compounds initially exist in the matrix in a single tautomeric form (1H). For 5M-6AzI it was found that it isomerizes under UV radiation yielding 3H-5M-6AzI and 6H-5M-6AzI. The formation of 6H-5M-6AzI was only observed in the argon matrix, and by intermediacy of 5M-6AzI-yl radical. 5M-4AzI also isomerizes to 3H-5M-4AzI. However, the formation of the 5M-4AzI-yl radical or the 4H-5M-4AzI tautomer was not observed. The different dependence on the host material shows that the tautomeric reactions observed should be governed by two distinct mechanisms, one involving radicals and depending on the host material, and another is not.Attachment of methoxy group to position 5 of the benzimidazole ring allowed tautomers 1H-5M-BzIm and 3H-5M-BzIm to be distinguishable. Each of them might exist in two distinct conformations (syn and anti) depending on the orientation of the methoxy group relatively to the ring, resulting in four minima on the potential energy surface of 5M-BzIm, all of them with close energies. The crystal structure of 5M-BzIm was resolved by X-ray diffraction, consisting of only the 3H-5M-BzIm tautomer, and in only one conformation (anti). However, in the gas phase and in the cryogenic matrices, the four most stable species of this molecule coexist, with populations close to the theoretically predicted. The individual signatures of each species present in the xenon matrix were spectroscopically assigned. For each tautomer, it was verified that the conformational populations could be manipulated by monochromatic vibrational excitation, by the exposure to the spectrometer light source, or thermally.For the unique conformer of 5M-4AzI isolated in a Xe matrix at 10 K, was observed its conformational isomerization, when exposed to monochromatic IR radiation. Besides, it was observed that this new high energy conformer totally relaxes, when exposed to the light source of the spectrometer, or thermally (T= 39 K). The rationalization of the observed results was supported by computation calculations at the B97-1/Def2-TZVP and MP2/Def2-TZVP levels of theory. The detailed study of the potential energy surfaces of 5M-4AzI and 5M BzIm allowed for a comparative analysis and explained the reason of higher difficulty in production and stabilization of the higher energy conformer in the case of 5M-4AzI, comparing with the conformers of high energy of 5M-BzIm.
