dc.description.abstractA formação assimétrica de ligações carbono-carbono usando ligandos quirais, é atualmente um dos temas mais estudados na síntese assimétrica. A obtenção de compostos enantiomericamente puros, é, cada vez mais, de extrema importância, para que estes sejam reconhecidos pelo ambiente quiral onde são inseridos. A alquilação enantiosseletiva de aldeídos e as reações assimétricas de Henry, são reações de formação de ligações C-C, obtendo-se álcoois secundários quirais através da reação de aldeídos com compostos organometálicos. Os álcoois secundários quirais são percursores na síntese dos mais variados compostos biologicamente ativos, nomeadamente, fármacos, perfumes, agroquímicos e muitos outros grupos funcionais. Neste projeto, foram desenvolvidos novos ligandos quirais com funcionalidades adequados à utilização em reações de catálise assimétrica. Esses ligandos foram ensaiados em reações de alquilação enantiosseletiva de aldeídos, bem como em reações assimétricas de Henry.Na primeira parte desta monografia, encontram-se descritos os processos de síntese dos diversos ligandos quirais derivados do ácido canforsulfónico, ácido (1R,3S)-canfórico e da L-cisteína. Os ligandos obtidos possuem funcionalidade diversificada como, sulfonamidas, hidroxi-sulfonamidas, imino-sulfonamidas, amino-sulfonamidas, diaminas, diiminas, entre outras.Foi sintetizada uma série de sulfonamidas derivadas dos ácidos (R)-(-)-10- e (S)-(+)-10-canforsulfónico, por reação dos cloretos de ácido respetivos com diferentes aminas. As sulfonamidas foram reduzidas às hidroxi-sulfonamidas respetivas, de modo a avaliar o efeito da presença do grupo hidroxilo nas reações de alquilação enantiosseletiva. A preparação de ligandos do tipo amino- e imino-sulfonamida, mostrou ser um processo bastante moroso, sendo necessário efetuar várias tentativas de síntese, de modo a obter bons rendimentos. Estes ligandos foram preparados por reação do grupo carbonilo da sulfonamida N-benzil-1-((1R,4S)-7,7-dimetil-2-oxobiciclo[2.2.1]heptan-1-il)metanosulfonamida com benzilamina e fenil-hidrazina, seguida de redução das iminas respetivas para obter as amino-sulfonamidas.Foram também preparados ligandos derivados do ácido canfórico contendo grupos imina e amina. Para tal, partiu-se do (1R,3S)-1,3-diamino-1,2,2-trimetilciclopentano, que se fez reagir com 2,5-diclorobenzaldeído e o-metoxibenzaldeído. A irradiação por micro-ondas mostrou ser um método rápido e eficaz para a síntese destes ligandos, com tempos de reação de 15 minutos e bons rendimentos. As diiminas foram reduzidas às diaminas correspondentes usando boro-hidreto de sódio, como agente redutor.Foi ainda sintetizada uma série de tiazolidinas, com diferentes grupos substituintes nas posições C2 e C4 do anel, com maior ou menor impedimento estereoquímico, de modo a avaliar este tipo de ligandos heterocíclicos nas reações assimétricas de Henry. Partindo da L-cisteína ou do seu éster metílico, foram sintetizados vários ligandos com diferentes substituintes em C2 por reação com diferentes aldeídos. A redução do grupo éster em C4 da (R)-2,2-dietil-1,3-tiazolidina-4-carboxilato de metilo, ou a reação do ácido com (R)- e (S)-feniletilamina, permitiu sintetizar o álcool ou as amidas respetivas.A reação de alquilação enantiosseletiva de aldeídos com dietilzinco, usando benzaldeído como substrato padrão, foi estudada com os ligandos quirais sintetizados a partir dos ácidos (R)-(-)-10- e (S)-(+)-10-canforsulfónico. O ligando N-benzil-1-((1R,4R)-2-(benzilamina)-7-7-dimetilbiciclo[2.2.1]hepan-1-il) metanosulfonamida permitiu obter os melhores resultados: 85% de conversão, 86% de produto quiral e um er de 24:76 (R:S), usando ciclo-hexano como solvente e 15 mol% de ligando, à temperatura ambiente. As reações de alquilação a temperaturas mais baixas, 0 ºC, mostraram conduzir a piores resultados neste processo catalítico. A presença de grupos mais volumosos, em substituição do grupo carbonilo do sistema bicíclico, mostrou estar associada a melhores resultados na reação de alquilação enantiosseletiva de aldeídos.Posteriormente, e utilizando o melhor ligando, foram otimizados alguns parâmetros da reação, nomeadamente o tipo de solvente e percentagem de ligando. Conclui-se que utilizando tolueno como solvente, embora a conversão fosse ligeiramente inferior (82%), a reação de catálise conduzia a um melhor er, 19:81 (R:S). Verificou-se que a percentagem de ligando que permitia obter melhores resultados era 15 mol%, quando se usava tolueno como solvente. Por fim, de modo a avaliar a abrangência da reação, estendeu-se a reação de catálise a outros substratos. Foram usados vários aldeídos aromáticos, com grupos dadores e atratores de eletrões, e alifáticos, tendo-se verificado que o melhor resultado foi obtido com o o-metoxibenzaldeído, que apresentou uma conversão >99% e um er de 10:90 (R:S).por
dc.description.abstractThe asymmetric formation of carbon-carbon bonds, using chiral ligands, is currently one of the most studied topics in asymmetric synthesis. Obtaining enantiomerically pure compounds is increasingly of utmost importance, so that they may be recognized by the chiral environment where they are inserted. The enantioselective alkylation of aldehydes and the asymmetric Henry reactions are C-C bond formation reactions, which originate secondary chiral alcohols through the reaction of aldehydes with organometallic compounds. Secondary chiral alcohols are precursors in the synthesis of a great diversity of biologically active compounds, namely, pharmaceuticals, perfumes, agrochemicals and many other functional groups. In this project, we developed new chiral ligands suitable for asymmetric catalysis reactions. These ligands were tested in enantioselective alkylation of aldehydes, as well as in asymmetric Henry reactions.In the first part of this thesis, the processes for the synthesis of various chiral ligands derived from camphorsulfonic acid, (1R,3S)-camphoric acid and L-cysteine are described. The resulting ligands have various functionalities, such as sulphonamides, hydroxysulfonamides, iminosulphonamides, aminosulphonamides, diamines and diimines, among others.A series of sulphonamides derived from (R)-(-)-10- and (S)-(+)-10-camphorsulfonic acids were synthetized by reaction of the respective acid chlorides with different amines. The sulphonamides were reduced to the corresponding hydroxysulfonamides in order to evaluate the effect of the presence of the hydroxyl group on the enantioselective alkylation reactions. Obtaining ligands of the amino- and imino-sulphonamide type proved to be a time-consuming process, requiring several attempts at the synthesis before it was possible to obtain the products in good yields. These ligands resulted from the reaction of the carbonyl group of N-benzyl-1-((1R,4S)-7,7-dimethyl-2-oxobicyclo[2.2.1]heptan-1-yl) methanesulfonamide with benzylamine and phenyl-hydrazine, followed by reduction of the resulting imines to obtain the amino-sulfonamides.Ligands derived from camphoric acid, containing imine and amine groups, were also prepared. In order to obtain these ligands, we started with (1R,3S)-diaminocyclopentane and reacted it with 2,5-dichlorobenzaldehyde and o-methoxybenzaldehyde. Microwave irradiation proved to be a fast and efficient method for the synthesis of these ligands, with reaction times of about 15 minutes and good yields. The diimines were reduced to the corresponding diamines using sodium borohydride.A series of thiazolidines having different substituent groups at C2 and C4 of the heterocycle were also synthesized, with more or less steric hindrance, in order to evaluate this type of heterocyclic ligand in the asymmetric Henry reactions. Starting from L-cysteine or its methyl ester, several ligands with different C2 substituents were synthesized by reaction with different aldehydes. Reduction of the ester group of methyl (R)-2-2-diethyl-1,3-thiazolidine-4-carboxylate, or reaction of the acid with (R)- and (S)-phenylethylamine, allowed the synthesis of the alcohol or amides.The enantioselective alkylation of aldehydes with diethylzinc using benzaldehyde as the standard substrate, was studied with the chiral ligands synthesized from (R)-(-)-10- and (S)-(+)-10-camphorsulfonic acid. The N-benzyl-1-((1R,4R)-2-(benzylamine)-7-dimethylbicyclo[2.2.1]hepan-1yl) methanesulfonamide ligand gave the best results: 85% conversion, 86% chiral product and an er of 24:76 (R:S), using cyclohexane as solvent and 15 mol% ligand, at room temperature. The alkylation reactions at lower temperatures, 0 °C, led to worse results in this catalytic process. The presence of more bulky groups, replacing the carbonyl group of the bicyclic system, proved to be associated with better results in the enantioselective alkylation of benzaldehyde.Subsequently, and using the best ligand, some parameters of the reaction were optimized, namely the type of solvent and percentage of ligand. We concluded that using toluene as solvent, although the conversion was slightly lower (82%), the catalysis reaction led to a better er, 19:81 (R:S). It was found that the percentage of ligand that allowed better results was 15 mol%, when the solvent of the reaction was toluene. Finally, in order to evaluate the scope of the reaction we extended the catalyst reaction to other substrates. A number of aromatic aldehydes with electron-donating and eletron-withdrawing substituents and aliphatic groups were used. The best result was obtained with o-methoxybenzaldehyde, which had a conversion >99% and an er of 10:90 (R:S).eng
