Abstract: A obtenção de compostos enantiomericamente puros tem uma procura cada vez maior, tendo a catálise assimétrica sido a técnica mais usada para a obtenção exclusiva de produtos enantiomericamente puros a partir de reagentes aquirais. Uma das reações mais importantes de catálise assimétrica é a alquilação enantiosseletiva de aldeídos com dietilzinco na presença de um ligando quiral, que permite obter álcoois secundários oticamente ativos. O trabalho descrito nesta dissertação prende-se com a síntese de ligandos quirais e a sua utilização como catalisadores na alquilação enantiosseletiva de aldeídos com dietilzinco para dar origem a álcoois secundários quirais.Na primeira parte do trabalho descrito nesta dissertação, efetuaram-se vários estudos sintéticos visando a obtenção de novos ligandos quirais. Foram utilizados como materiais de partida o ácido L-glutámico e a L-cisteína, aminoácidos de fonte quiral natural. A partir do glutamato de dimetilo e por irradiação de micro-ondas, foi obtida uma ϒ-lactama quiral, que foi ensaiado na alquilação enantiosseletiva do benzaldeído com dietilzinco, demostrando ser pouco eficiente.Utilizando a L-cisteína e o seu éster metílico, sintetizou-se uma diversidade de novas tiazolidinas. Foi desenvolvido um método sintético, usando irradiação de micro-ondas, que permitiu a metilação eficiente do azoto da tiazolidina, originando heterociclos N-metilados com grupos carboxilo, éster e hidroximetílico em C4. Utilizando uma via sintética simples foi possível obter vários compostos tiazolidínicos substituídos em C2, por reação do hidrocloreto do éster metílico da L-cisteína com vários aldeídos aromáticos, possuindo grupos atratores e dadores de eletrões. Os grupos éster em C4 das tiazolidinas resultantes foram subsequentemente reduzidos, utilizando como agente redutor NaBH4/CaCl2, às tiazolidinas 4-hidroximetílicas correspondentes.Foram ainda sintetizadas tiazolidinas com outros grupos funcionais em C4, nomeadamente grupos imina e amina, através de sequências sintéticas de vários passos. Partindo da 4-hidroximetiltiazolidina, foi efetuada a proteção do azoto, tosilação do grupo hidroxilo em C4, formação da azida e hidrogenólise desta a amina. Esta amina, por reação com o salicilaldeído, deu origem à imina, que posteriormente foi reduzida à amina correspondente. Foi ainda efetuada a reação da 4-tosiloximetiltiazolidina com ciclo-hexilamina com o objetivo de obter uma nova tiazolidina, com um grupo ciclo-hexilamina em C4. Inesperadamente, desta reação resultou um produto diferente, um composto bicíclico resultante de uma ciclização intramolecular, devido a uma reatividade diferente da habitual do grupo tosiloxi.Na segunda parte do trabalho descrito nesta dissertação, tiazolidinas N-metiladas e tiazolidinas 4-hidroximetílicas. Os ligandos N-metilados revelaram-se pouco eficientes e seletivos, tendo o melhor resultado sido obtido na presença da tiazolidina N-metilada, com o grupo éster em C4, que apresentou uma conversão de 99% e um ee de 27%, para o (S)-1-fenilpropanol.Utilizando o melhor ligando, a tiazolidina com o grupo 3-clorofenilo em C2, foram realizados estudos de otimização com diferentes solventes, concluindo-se que o ciclo-hexano era o melhor solvente para este tipo de ligando. De modo a avaliar a abrangência do ligando, foram ensaiadas reações de alquilação com diferentes aldeídos aromáticos e alifáticos. Com os substratos usados obteve-se uma variedade de conversões, desde completas até inferiores a 10%. Relativamente aos excessos enantioméricos, estes foram moderados a baixos e em todos os casos inferiores ao obtido na alquilação do benzaldeído. A seletividade mais elevada foi observada quando foram usados como substratos o o-anisaldeído e o 2-naftaldeído, que originaram produtos com ee de 57% e 60%, respetivamente.
There is presently an increasing demand for obtaining enantiomerically pure compounds. Asymmetric catalysis has been the most frequently used technique for the exclusive synthesis of optically pure products from achiral reagents. One of the most important reactions of asymmetric catalysis is the enantioselective alkylation of aldehydes with diethylzinc in the presence of a chiral ligand, which allows the formation of optically active secondary alcohols. The work described in this dissertation deals with the synthesis of chiral ligands and their application as catalysts in the enantioselective alkylation of aldehydes with diethylzinc to give chiral secondary alcohols.In the first part of the work described in this dissertation, several synthetic studies were carried out in order to obtain new chiral ligands. L-glutamic acid and L-cysteine, natural chiral source amino acids, were used as starting materials. Starting from dimethyl glutamate and using microwave irradiation, a chiral ϒ -lactam was obtained, which was tested in the enantioselective alkylation of benzaldehyde with diethylzinc, proving to be inefficient.Using L-cysteine and its methyl ester a variety of novel thiazolidines was synthesized. A synthetic method was developed using microwave irradiation, which allowed efficient methylation of the thiazolidine nitrogen, originating N-methylated heterocycles with carboxyl, ester and hydroxymethyl groups at C4.Using a simple synthetic route it was possible to obtain various C2-substituted thiazolidines through the reaction of L-cysteine methyl ester hydrochloride with various aromatic aldehydes, having electron attracting and electron donating substituents. The C4 ester groups of the resulting thiazolidines were subsequently reduced, using the NaBH4 / CaCl2 as reducing agent, to the corresponding 4-hydroxymethyl thiazolidines.Thiazolidines with other functional groups at C4, in particular imine and amine groups, were also synthesized through multi-step synthetic sequences. Starting from 4-hydroxymethylthiazolidine, protection of the nitrogen, tosylation of the hydroxyl group at C4, formation of the azide and its hydrogenolysis to the corresponding amine were carried out. This amine, upon reaction with the salicylaldehyde, gave the imine, which was subsequently reduced to the corresponding amine.The reaction of the 4-tosyloxymethylthiazolidine with cyclohexylamine was also carried out in order to obtain a new thiazolidine with a cyclohexylamine group at C4. Unexpectedly, this reaction resulted in a different product, a bicyclic compound resulting from an intramolecular cyclization, due to a different reactivity than usual of the tosyloxy group.In the second part of the work described in this dissertation, the various synthesized ligands, N-methylated thiazolidines and 4-hydroxymethyl thiazolidines, were evaluated as chiral ligands in the enantioselective alkylation of benzaldehyde with diethylzinc, using previously optimized conditions. The N-methylated ligands were shown to be poorly efficient and selective, and the best result was obtained in the presence of the N-methylated thiazolidine with the ester group at C4, which showed a conversion of 99% to(S) -1-phenylpropanol, with an ee of 27%.The ligands with the hydroxymethyl group at C4 and substituted at C2 with aromatic groups, proved to be efficient in the alkylation reaction of benzaldehyde with diethylzinc, with conversions greater than 90% being obtained. The most selective ligands were the thiazolidines with the 2-chlorophenyl and 3-chlorophenyl groups at C2, giving products with enantiomeric excesses of 61% and 67%, respectively. In all cases, the (S) -enantiomer of 1-phenylpropanol was always obtained.Using the best ligand, the thiazolidine with the 3-chlorophenyl group at C2, optimization studies with different solvents were carried out, concluding that cyclohexane was the best solvent for this type of ligand. In order to evaluate the efficiency of the ligand, alkylation reactions were carried out with different aromatic and aliphatic aldehydes. With the substrates used, a variety of conversions were obtained, from complete to less than 10%. The enantiomeric excesses were moderate to low and in every case lower than the ee obtained in the alkylation of benzaldehyde. The highest selectivity was observed when o-anisaldehyde and 2-naphthaldehyde were used as substrates, yielding products with 57% and 60% ee, respectively.
