Title: Efeitos do exercício prolongado com e sem impacto axial em lesões na barreira hematoencefálica e em marcadores periféricos de fadiga central

Abstract: A síntese das monoaminas e das catecolaminas no cérebro possuem papel muito importante durante o exercício prolongado. Nesse caso, a maior atividade do sistema serotoninérgico tem sido ligada à instalação da fadiga central e, atualmente, reconhece-se que a barreira hematoencefálica desempenha um papel chave na interação entre o transporte dos precursores e a síntese do neurotransmissor. Estudos já apontaram também que a estrutura da barreira hematoencefálica pode ser comprometida durante a prática de alguns exercícios, porém, ainda é necessário saber se estas alterações são capazes de modular a atividade do sistema serotoninérgico durante a atividade. O objetivo deste trabalho é verificar as associações existentes entre lesões na barreira hematoencefálica e marcadores periféricos de fadiga central, induzidas por exercício prolongado. Metodologia: oito triatletas treinados (n=8) do sexo masculino, com idades de 19,87±2,29 anos, realizaram dois protocolos na mesma intensidade relativa de esforço. O exercício contínuo, prolongado (40 minutos) foi realizado em intensidade estipulada em 75% do VO2máx, no primeiro momento em cicloergômetro e, em outro momento, em corrida. Foram coletadas amostras de sangue e saliva, antes e após cada protocolo, para análise de S100-B, Prolactina, Cortisol e Deidroepriandrosterona (DHEA). Resultados: o valor médio do hormônio Cortisol detectado na saliva foi maior após a realização do exercício de ciclismo relativamente aos valores médios basais, porém, esta diferença não foi significativa (p=0,123), sendo que os valores pós-corrida para o mesmo hormônio permaneceram inalterados comparativamente ao repouso. O DHEA exibiu um aumento significativo após os exercícios, tanto em cicloergômetro (p=0,012) quanto em corrida (p=0,025). A análise de S100-B no soro não demonstrou alterações significativas em nenhuma das situações experimentais. No entanto, os índices de prolactina aumentaram expressivamente após corrida (p=0,012), e um sutil e insignificante aumento foi observado após ciclismo (p=0,123). Conclusão: os índices de S100-B, marcador de lesão na barreira hematoencefálica, não foi diferente entre as atividades que envolveram impacto axial (corrida) da atividade de ciclismo (sem impacto). Nesse sentido, sugerimos que o protocolo utilizado (principalmente em corrida) produziu efeitos significativos nos marcadores de fadiga central, sem interferir na permeabilidade da barreira hematoencefálica. The synthesis of monoamines and catecholamine in the brain have very important role during prolonged exercise. In this case, the major activity of the serotonergic system has been connected with the phenomenon of central fatigue and, currently, it is acknowledged that the blood-brain barrier plays a key role in the interaction between the transport of precursors and the synthesis of the neurotransmitter. Studies have also pointed out that the structure of the blood-brain barrier can be compromised during the practice of some exercises, however, it is still necessary to know if these changes are able to modulate the activity of the serotonergic system during activity. The aim of this study is to verify the existing associations between blood-brain barrier leakage and peripheral markers of central fatigue induced by prolonged exercise. Methods: eight male trained triathletes (n = 8) with a mean age of 19.87±2.29 years conducted two protocols in the same relative intensity of effort. The continuous, prolonged exercise (40 minutes) was conducted in 75% of VO2max in cycle ergometer at first and, later in the run. Blood and saliva samples were collected, before and after each protocol for examination of S100-B, Prolactin, Cortisol and DHEA. Results: the mean value of the salivary cortisol was greater after the cycling exercise in relation to the average basal values, however, this difference was not significant (p = 0.123), post-race values for the same hormone remained unchanged compared to baseline. DHEA exhibited a significant increase after the both exercises, in cycle ergometer (p = 0.012) and race (p = 0.025). The analysis of S100-B in serum did not show significant changes in any of the experimental situations. However, prolactin increased significantly after the race (p = 0.012), and a subtle and insignificant increase was observed after cycling (p = 0.123). Conclusion: the S100-B (injury marker in the blood-brain barrier), was not different between the exercises involving axial impact (run) and cycling (no impact). Accordingly, we suggest that the protocol used (mainly in run) produced significant effects on markers of central fatigue, without interfering in the permeability of the blood-brain barrier.
