Abstract: Nos últimos anos, o crescimento do conhecimento acerca dos polímeros inteligentes tem aberto novas portas na biotecnologia, medicina e engenharia. Estes polímeros são formalmente conhecidos pela sua capacidade de responder, de uma forma reversível, a estímulos provocados no meio ambiente. Entre esses estímulos, os mais estudados são as mudanças de temperatura e de pH, os quais podem provocar variações na estrutura, características da superfície, solubilidade, transição sol-gel, entre outras especificações dos polímeros. Na síntese destes polímeros, a técnica de CLRP é a mais utilizada pois permite obter polímeros e copolímeros com massas moleculares controladas bem como um baixo índice de polidispersividade, permitindo obter uma resposta fina e controlada. Quando incluídos em sistemas copoliméricos, este tipo de comportamento dos polímeros inteligentes desempenha um papel importante nos processos de self-assembly. Na resposta a uma variação do meio, as macromoléculas podem formar nanoestruturas estáveis e bem definidas. A predeterminação destas nanoestruturas com base no desenho molecular das estruturas moleculares é um passo importante no avanço da modelação da síntese polimérica de modo a obter as estruturas desejadas. O copolímero de bloco utilizado como caso de estudo foi preparado com segmentos de um polímero hidrofílico, PEG, e de um polímero que apresenta resposta ao pH, P4VP. A P4VP é um polielectrólito fraco com pKa aproximadamente de 4,5. Quando o valor de pH é superior ao valor do pKa, a P4VP apresenta uma mudança na sua hidrofilicidade, passando a ser um segmento hidrofóbico. Pretende-se com este estudo, relacionar a estrutura que os polímeros inteligentes formam com as suas propriedades físicas e químicas quando estimulados ambientalmente, nomeadamente através da variação do pH do meio. Com o objectivo de encontrar um equilíbrio e polaridade adequada para o self-assembly recorreu-se ao copolímero de PEGx-P4VPy com diferentes comprimentos de cada segmento de polímero. As características físicas e químicas e a estrutura destes copolímeros foram seguidas por métodos reológicos e por métodos de DLS, respectivamente. Verificou-se que a P4VP promove três comportamentos diferentes quando ocorre variação do pH: viscosidade inicial associada a uma estrutura distendida, devido às interacções das cargas positivas do polímero com a água; aumento de viscosidade, associada à formação micelar; e viscosidade constante, associada à existência estável das estruturas micelares. Estes resultados foram verificados com o estudo por DLS, onde se verificou que, a partir do copolímero mPEG-b-P4VP se podem formar estruturas estáveis e de pequenas dimensões, e que o tipo de estruturas formadas depende, não só das condições do meio como do desenho molecular dos copolímeros (composição e peso molecular). Este aumento da área superficial das cargas expostas às interacções com a água pode relacionar-se com o aumento da viscosidade. O tipo de nanoestruturas formadas, para um dado copolímero a um dado pH, é indicada pela determinação de variáveis físicas das dispersões do copolímero, tais como a transparência, a viscosidade e o volume hidrodinâmico. O controlo do tipo de estruturas é conseguido através do balanço entre o comprimento de bloco de PEG e P4VP e a concentração do copolímero em solução. Verifica-se que a utilização de copolímeros de bloco com maior equilíbrio entre os segmentos hidrofílicos e hidrofóbicos e de menor peso molecular favorece a formação de nanoestruturas micelares esféricas e estáveis por self-assembly ocorre.
In recent years, the knowledge about the smart polymers has growth and has open new doors in biotechnology, medicine and engineering. These polymers are known for their ability to be triggered in a reversible way to the stimulus caused in the environment. Among these stimuli, the most studied are the changes in the environmental temperature and pH, which may cause variations in their structure, surface characteristics, solubility, sol-gel transition, among other specifications of polymers. In the synthesis of these polymers, CLRP technique is the most used since it gives polymers and copolymers with controlled molecular weights and a low polydispersity index, to allow a fine and controlled trigerring. When these type of polymers is include in copolymeric systems, their responsive behavior can play a critical role on self-assembly mechanisms. When triggered by an environmental stimulus, these systems can form stable and well defined nanostructures. The predetermination of these characteristics on the basis of the copolymers molecular design is an important step in the advancement of modeling the polymer synthesis to obtain the desired structures. The block copolymer used was prepared with hydrophilic polymer segments of PEG and hydrophobic of P4VP. The P4VP is a weak polyelectrolyte with pKa approximately 4.5. When this value is exceeded, the P4VP presents a change in their hydrophilicity. The aim of this study was to relate the structure of the smart polymers with their physical and chemical properties when triggered environmentally, namely by varying the pH. With the aim of finding a proper polarity for the self-assembly processed, we evaluated the use of PEGx-P4VPy block copolymers with different lengths of each segment polymer. The physical and chemical structure of these copolymers was followed by rheological methods and by DLS analysis. It was found that P4VP presents three different behaviors with the pH variation: initial viscosity associated to an extended structure due to the interaction of the positive charges of the polymer with water; increase of viscosity associated to micelle formation and; constant viscosity, associated with the stability of micellar nanostructure formed. These results were verified with the DLS study that show the formation of stable and nano structures. This increase in surface area expose the positive charge to interactions with the water, associated with the increase in viscosity. The type of nanostrutures that are formed, for a given copolymer at a specific pH, is indicated by the determination of physical variables of the copolymers aqueous dispersions, such as the transparency, the viscosity and the hydrodynamic volume. The control of the nanostruturation is accomplished by the balance between block length of PEG and P4VP and the concentration of the P4VP into copolymer solution. It is noted that the use of block copolymers with greater balance between the hydrophobic and hydrophilic segments and lower molecular weight promotes the formation of nanostructures and stable spherical micelle by self-assembly.
